Whole-genome profiling in Brassica genetic research and breeding

چکیده
Whole-genome profiling techniques have been in use for two decades to develop genetic maps and to understand genetic diversity in a wide range of agriculturally important plant and animal species, including Brassica crop species. The genetic mapping approach has been useful in developing and validating markers linked to major gene traits which have then been used as diagnostic markers in marker-assisted selection programmes. An extension of the linkage mapping approach – Quantitative Trait Locus (QTL) mapping – has been widely used to identify large chromosomal segments associated with quantitatively expressed traits (controlled by major and/or minor genes) in controlled cross populations. New whole-genome profiling tools are becoming available in many crop and livestock species and promise to improve our understanding of genetic diversity and the genetic control of agronomic traits. In this review, we discuss past, present and future whole-genome profiling tools in Brassica; applications of whole-genome profiling for Brassica genetic research; and anticipated applications in Brassica breeding programmes.

Background Methods for haplotyping and DNA copynumber typing of single cells are paramount for studying genomic heterogeneity and enabling genetic diagnosis. Before analyzing the DNA of a single cell by microarray or next-generation sequencing, a whole-genome amplification (WGA) process is required, but it substantially distorts the frequency and composition of the cell’s alleles. As a conseque...

متن کاملUnveiling the genetic loci for a panicle developmental trait using genome-wide association study in rice
Panicle size has a high correlation with grain yield in rice. There is a bottleneck to identify the additional quantitative trait loci (QTL) for panicle size due to the conventional traits used for QTL mapping. To identify more genetic loci for panicle size, a panicle developmental trait (LNTB, the length from panicle neck-knot to the first primary branch in the rachis) related to panicle size ...
متن کاملSpecies-wide genome sequence and nucleotide polymorphisms from the model allopolyploid plant Brassica napus
Brassica napus (oilseed rape, canola) is one of the world's most important sources of vegetable oil for human nutrition and biofuel, and also a model species for studies investigating the evolutionary consequences of polyploidisation. Strong bottlenecks during its recent origin from interspecific hybridisation, and subsequently through intensive artificial selection, have severely depleted the ...
متن کاملI-37: Establishing High Resolution Genomic Profiles of Single Cells Using Microarray and Next-Generation Sequencing Technologies
The nature and pace of genome mutation is largely unknown. Standard methods to investigate DNA-mutation rely on arraying or sequencing DNA from a population of cells, hence the genetic composition of individual cells is lost and de novo mutation in cell(s) is concealed within the bulk signal. We developed methods based on (SNP-) arraying and next-generation sequencing of single-cell whole-genom...
